In vitro response of Cerastium candidissimum nodal explants to three types of cytokinins and NAA

Abstract
The application of tissue culture for the Greek endemic plant, Cerastium candidissimum, would be valuable for commercial and conservation purposes.
This study aims to explore how the in vitro multiplication of this species is affected by three different cytokinins, i.e., zeatin (ZEA), kinetin (KIN), and 2-isopentenyl adenine (2iP), with and without the addition of 1-naphthaleneacetic acid (NAA). Nodal explants excised from in vitro grown seedlings were cultured on solid (8 g L‑1 agar), Murashige and Skoog (MS) media, supplemented with 30 g L‑1 sucrose.
The cytokinins ZEA, KIN, or 2iP were used at 0.5 mg L‑1 coupled or not with the auxin NAA at 0.1 mg L‑1. Media with no plant growth regulators (Hf) served as a control.
A completely randomized, two-factorial design was used, i.e., population (Mount Parnassos (PS), mt.
Parnitha (PR), mt.
Hymettus (HM)) × medium (Hf, ZEA/NAA, KIN/NAA, or 2iP/NAA). Both population and medium had a significant effect on total shoot number and node number, while no effect was observed on shooting percentage which was 89.2-95.4% of all three populations.
Explants originating in Mount Parnassos (PS) produced the highest shoot number (2.7) as well as the highest node number (3.4). The two main factors had a significant interaction on the shoot length, and it was registered that the longest shoots (5.3 cm) were produced in Hf media or media containing 0.5 KIN/0.1 NAA (4.2 cm) by explants coming from the PR population.
Nonetheless, spontaneous rooting of explants in all media and of all origins was observed.
Rooted plantlets were successfully acclimatized (90-100%) in peat:perlite 1:1 (v/v) and the final survival percentage was 100%, 30 days after accomplishing acclimatization.
