Micropropagation of Hypericum taygeteum, an endemic and rare aromatic species of Greece

Abstract
Hypericum taygeteum (Hypericaceae) is an endangered small aromatic and pharmaceutical perennial herb of Greece.
It is endemic to three restricted and small areas in southern Peloponnese, and it typically grows in dry habitats at crevices of shaded limestone rocks.
The present study was focused on micropropagation of this species.
Nodal shoots were mainly collected from native adult plants at Langada gorge of Taygetus mountain and used as an explant source.
After surface disinfection, the explants were inoculated on half or full-strength MS media containing either 1.5 or 3% sucrose.
Petri dishes were used as vessel cultures covered with commercial semipermeable membranes or parafilm.
The higher rate of proliferation was achieved when explants from the middle and the lower part of the stem taken from the mother plant were chosen and cultured on half-strength MS substrates containing 1.5% sucrose.
Seasonal variations in the regeneration of explants were observed; explants collected in May gave the highest multiplication rate compared to those collected in June, August, and October.
Higher proliferation rates, and simultaneous rooting of microshoots, were achieved when the explants in the proliferation stage remained on the substrates for longer than 60 days.
Covering the Petri dishes with a semi-permeable membrane increased the proliferation rate at all stages of micropropagation and increased the acclimatization rate (up to 100%) of plantlets in ex vitro conditions.
The plantlets transplanted in the soil showed normal growth without any morphological variation.
