Establishment of two different suspension cell lines in Chinese jujube (Ziziphus jujuba Mill.)

Abstract
Taking the embryonic callus of ‘Wuhezao76’ and pulp callus of ‘Fuxiangzao’ as experimental materials, two stable suspension cell cultivating systems were successfully established.
The results showed that the medium containing MS + maltose 20 g L-1 + NAA 0.5 mg L-1 + TDZ 3.0 mg L-1 was suitable for both the initiation and proliferation cultivation of jujube embryonic callus suspension cells.
The medium containing MS + sucrose 30 g L-1 + NAA 0.5 mg L-1 + TDZ 3.0 mg L-1 was suitable for establishing jujube pulp callus suspension cells.
Both stable suspension cell lines could be established after 2 months of cultivation by removing large cell clusters every 7 days.
The growth curve of the suspension cell lines showed an “S” shape, and the cells grew best at 2-4 days after changing the fresh medium.
The results of this study can provide a material basis for further research on the genetic transformation, efficient production of secondary metabolites, and metabolic regulation in Chinese jujube.
