Identifying Cucumber mosaic virus (Cucumovirus, Bromoviridae) isolates on Impatiens (Balsaminaceae): a preliminary study using molecular characterization

Abstract
Impatiens is a genus of plants with several species native to Indonesia and is known for its ornamental value. Impatiens is also known to be a host for several plant viruses making its propagation for commercial purposes challenging. Cucumber mosaic virus (CMV) is a plant pathogen commonly found in a wide host range, including vegetables, fruits, and ornamental plants, including Impatiens. CMV causes mosaic pattern and malformation in leaves, and discoloration of flowers.
The research aimed to identify the virus infecting Impatiens with the mosaic symptom using molecular techniques.
The study was carried out by a collection of symptomatic indicator plants (Solanum lycopersicum) as a result of inoculation with the mosaic virus from the Impatiens plant (previous research results), extraction of total RNA, quantification and qualification of total RNA, RT-PCR using specific primer pair of CMV, and sequencing analysis.
The results showed that CMV was successfully amplified from the test plants.
RT-PCR using specific primer pair successfully amplified the virus target on 600 bp.
Nucleotide sequence analyses showed that CMV isolate from our samples shared high similarity with other CMV isolates available on GenBank (97% for nucleotide and 100% for amino acid).
