Evaluation of the photosynthetic apparatus functioning in Lavandula angustifolia Mill. at different stages of post aseptic adaptation

Abstract
The article presents the results of the photosynthetic apparatus activity estimation in the leaf blades of Lavandula angustifolia cultivars made by the method of chlorophyll a fluorescence induction kinetics change.
Chlorophyll fluorescence was measured with a portable pulse fluorometer MINI-PAM II (Heinz Walz, Germany, 2017). Four cultivars were compared: ‘Belyanka’, ‘Prima’, ‘Record’, and ‘Sineva’. Photosynthetic activity in the leaves was studied after dark adaptation for 30 min.
The studies were made in vitro, as well as after adaptation in vivo for 15 days, 1 month, 2 months and on the mother plants cultured in open fields, which were used as a control.
Based on the experiment results it can be supposed that ‘Prima’ has higher adaptive capacity and the fastest acclimatization to post aseptic conditions of in vivo culture.
The effective photochemical quantum yield Y(II) in ‘Prima’ after 1 month of adaptation increased from 0.24 up to 0.43 a.u. fluorescence.
After 2 months, the coefficient of fluorescence decay (vitality index) Rfd in this cultivar increased by 3 times (from 1.22 to 3.65 a.u.). The lavender cultivar ‘Record’ demonstrated a similar rate of adaptation, as it had doubled the effective photochemical quantum yield over two months of adaptation (from 0.18 to 0.43 a.u.), and the viability index Rfd increased by 6.5 times (from 0.69 to 4.49 a.u.). We recommend these cultivars to produce L. angustifolia planting material in vitro.
