Abstract
Improvement of in vitro propagation of Dendrobium is a prerequisite to an effective propagation technology.
The main objective of the study was to establish callus proliferation method of Dendrobium hybrid CF22-58 and Dendrobium hybrid Dian Agrihorti using liquid culture system in airlift bioreactor.
The research was conducted at the Tissue Culture Laboratory IOCRI from January to December 2018. The study consisted of two experiments.
The first experiment involved various combinations of three media: 1) ½ Murashige and Skoog (MS) + 0.5 mg L‑1 thidiazuron (TDZ) + 0.1 mg L‑1 benzyl amino purine (BAP), 2) ½ MS + 0.3 mg L‑1 TDZ + 0.3 mg L‑1 BAP, or 3) ½ MS + 0.1 mg L‑1 TDZ + 0.5 mg L‑1 BAP) and two cultivars of Dendrobium hybrid CF22-58 and Dendrobium hybrid Dian Agrihorti as subplot.
The second experiment involved combining callus densities (5, 10, and 15 g 250 mL‑1 media) with various aeration levels (2.5, 5.0, and 7.5 vessel volume per minute (vvm)). We found that genotype, media, callus density, and aeration level have significant effects on callus proliferation in airlift bioreactor.
Callus proliferation of Dendrobium hybrid CF22-58 was better and faster than Dendrobium hybrid Dian Agrihorti. The best callus proliferation in Dendrobium hybrid CF22-58 was observed in medium with ½ MS + 0.5 mg L‑1TDZ + 0.1 mg L‑1BA, aeration rate of 5 vvm and callus density of 15 g 250 mL‑1 medium.
In this medium callus proliferation reached 332.8-356.7% with multiplication rate of 4.3-4.6, callus proliferation of 70-81%, and callus browning of 31-33% in the results of this study can be potentially applied in doing callus proliferation of other species of Dendrobium.
