Abstract
Vanda is a genus of slow-growing orchid plants.
Micropropagation technique using plant shoot tip generally leads to losing the mother plants.
Therefore, a fast and optimized method for cloning Vanda orchids without sacrificing the source plants is needed.
This research aimed to explore the potentials of rachis explants for micropropagation of Vanda orchids.
Rachis was collected from 20 clones of Vanda. Young rachis segments of Vanda inflorescences (2-5 cm long) were used as explants, and cultured on two MS medium treatments: the first medium was ½ MS + 0.75 mg L‑1 TDZ + 0.25 mg L‑1 BAP + 20 g L‑1 sucrose and the second medium was MS + 0.75 mg L‑1 TDZ + 0.25 mg L‑1 BAP + 20 g L‑1 sucrose.
Callus/shoot growth was successfully induced in the rachis segments of Vanda inflorescences.
The best medium for callus/shoot initiation was MS + 0.75 mg L‑1 TDZ + 0.25 mg L‑1 BAP + 20 g L‑1 sucrose.
The shortest period of callus/shoot formation was in M416.139A Vanda clone, i.e., 58 days after culture.
Shoots from this culture were used as explants for further mass propagation.
