De novo regeneration capacity of different explants of Japanese plum (Prunus salicina Lindl.)

Abstract
Somatic plum plants were regenerated from callus and seeds of plum (Prunus salicina Lindl.) ‘Victoria’. The de novo regeneration capacity was tested in sliced calli derived from the base of proliferation clusters and in 5 different sections of seeds (axis 1, axis 2, cotyledon 1, cotyledon 2, and cotyledon 3) extracted from open-pollinated fruits.
Regenerated shoots were obtained from culture media containing 1.5, 2, and 2.5 mg L‑1 of 6-benzyladenine (BA) and indolebutyric acid in which both sliced calli and seeds were cultured for 28 days.
Using this procedure, calli explants induced more shoots than seed explants; additionally, a higher amount of calli explants induced shoots when compared with seeds.
In seeds, the origin of regeneration was studied, and direct regeneration was observed to be higher at the concentration of 1.5 mg L‑1 of BA, whereas indirect regeneration was detected at a larger level at the concentrations of 2 and 2.5 mg L‑1 of BA. Both parts of the axis, as well as the part of the cotyledons that was closer to the axis (cotyledon 1), were the sections that showed a stronger reaction to the shoot-induction treatments.
