Abstract
In Dahlia crops two diseases caused by the bacteria Erwinia chrysanthemi and the Dahlia mosaic virus have been reported to limit the production.
Efficient chemical control does not exist for the diseases.
Selection based on symptoms is also inadequate.
Only sanitary selections can bring a solution to produce pathogen free plants.
Immunoenzymatic test (Standard ELISA and B.A.system) is carried out to detect bacteria and virus in cuttings and main stem of diseased plants.

To increase the efficiency of this method, investigations have been reported as follows: 1) Extracting buffer: to limit the effect of tannins and polyphenolic substances in plant tissues and evaluate the nicotine and urea content for the accuracy to detect bacteria and virus; 2) Detection in different tissues of the plant; 3) Conditions to increase the possibilities to detect bacteria and virus simultaneously in the same sample; 4) This paper describes the incorporation of Biotin/ Avidin system (B.A.-ELISA) into standard ELISA Technique in order to improve the detection of bacteria and virus in plants when the antigens level is low.
