Abstract
Leaf material from field or glasshouse grown material as well as tissue cultured plants is used during disease indexing.

All diagnostic analysis of plant viruses is carried out using the ELISA technique, variations of both double antibody-sandwich and F(ab1)2 ELISA being employed.
Low level infections are catered for using the controlled environments typical of tissue culture laboratories which can be tailored to provide suitable light and temperature levels for optimum viral multiplication.
In many cases virus infections are only detected after this treatment has occurred.
Viroid infections are detected using an adapted PAGE technique which can be carried out in one day, bacterial and fungal diseases are detected using selective nutrient media during the culture passages.
