Abstract
Chromosome counts and/or measurements of stoma length of plants regenerated via shoot-forming callus of A. scherzerianum showed little variation in ploidy.
Correspondingly, cytophotometric determinations of nuclear DNA content in caulogenic callus, even after repeated subculturing, revealed values mostly around 2C. However, increasing cytogenetic instability, loss of shoot-forming ability, and habituation was observed, when portions of callus lacking shoot initials were isolated and subcultured separately.
It appears that the organized shoots exert a stabilizing influence on the ploidy level of the surrounding callus tissues.
As long as a high level of organogenesis is maintained in the cultures, the risk of changes in ploidy is neglegible.
