Abstract
A technique combining liquid and agar-solidified layers of media has been applied to the micropropagation of hyacinths cv.
Carnegie.
Regeneration has been induced in one-leaf microcuttings.
Different concentrations of BA and kinetin in solid and liquid Murashige-Skoog media have been tested.
Variation in the response of micro-cuttings has been observed: some explants produced only shoots whereas others also developed roots or became swollen.
Both types of media gave a similar yield of adventitious plantlets, while regeneration of leaves and their lengths were greatest on explants cultivated in all combinations of BA. The mean number of bulbs produced per explant was higher in the double-phase medium.
The percent of plantlets with wet vitrification was higher on double-phase medium than on solid medium.
