DOUBLE-STRANDED RNA ANALYSIS AS A METHOD FOR DETECTING VIRUSES IN WOODY ORNAMENTALS

Abstract
The technique of double-stranded RNA (dsRNA) analysis was described, and used to detect virus and virus-like agents in woody ornamentals and herbaceous test plants.
After phenol extraction of tissue to isolate cellular nucleic acids, the dsRNA was selectively purified by binding to cellulose powder in 17% ethanolic buffer.
The dsRNA which eluted from the cellulose in ethanol-free buffer was electrophoresed in polyacrylamide gels.
Readily detectable amounts of dsRNA were found in six woody ornamentals with virus-like symptoms although little or no dsRNA was found in a further five with virus-like symptoms.
No dsRNA was detected in five apparently healthy woody ornamentals.
The diagnostic potential of dsRNA analysis was discussed.
