Abstract
Mutants of a sterile Kohleria hybrid have been induced in a micro-propagation system and selected at the regenerated plant level.
Bud formation and subsequent plant regeneration occured at high rate from internode segments cultivated on Nitsch’s medium supplemented with 1 mg/l kinetin and 0.5 mg/l IAA. For mutagenesis, internode explants, from in vitro-grown shoots, were treated with N-nitroso-N-methylurea (NMH) prior to placing them on bud induction medium.
Initially, a mutant with enhanced flowering capacity under low light conditions was isolated.
Further treatment of this mutant with NMH yielded one especially attractive type, among others, in which flowering under low light conditions was coupled with significantly shorter internodes and smaller leaves.
Both this and the other variant types remained phenotypically stable through several cycles of vegetative propagation, thus confirming that these are true mutants.
