IN VITRO CULTURE OF IMMATURE EMBRIOS AND COTYLEDONS OF JUGLANS REGIA L. MORPHOLOGICAL AND ANATOMICAL ANALYSES OF SOME REGENERANTS

Abstract
A study has been carried out with a breeding purpose on regeneration capability of "in vitro" cultured embryos with parts of cotyledons, intact cotyledons and endosperm tissue of the new type of the Bulgarian walnut variety Izvor 10 (J. regia L) with lateral fruit set, similar to the most famous American cultivars.
Explants have been cultured on several different nutrient media at a 16 hr. photoperiod.
Normal plants with well developed central root, stem and leaves have been obtained after 55 days of culturing the embryos with cotyledons on a medium consisting of MS mineral salts and tiamine HCl 1 mg/l, nicotinic acid 1 mg/l, biotin 0.1 mg/l, pyridoxine HCl 1 mg/l and calcium panthotenate 1 mg/l, followed by transferring to the same medium with 1 mg/l BAP in addition.

Data from the morphological cytological and hystological analyses of regenerants with developmental deviations obtained through direct embryogenesis from explants of embryos with cotyledons are presented.
