VIRUS DIAGNOSIS IN SUCKERS AND TISSUE CULTURES OF BANANA

Abstract
In this study, virus diagnosis has been carried out in banana suckers (mother stocks used in culture initiation) and micropropagated cultures using DAS (Double Antibody Sandwich) – ELISA for indexing of BBTV (2004, 2005, 2006, 2007 and 2008), BSV (2005, 2006, 2007 and 2008), CMV (2006, 2007 and 2008) and BBrMV (2004, 2005, 2006, 2007 and 2008). In the year 2004, BBTV was detected in 9.63% of suckers (mother stocks) and in subsequent years, no BBTV was detected in any of the suckers collected from field for culture initiation.
In micropropagated cultures, all four-virus incidences (0.25–1.45%) were observed in almost all the years tested.
Further, the Polymerase Chain Reaction (PCR) was standardized, and there was known sized specific amplified product observed in BBTV (250 bp) and BSV (221 bp) infected samples, which was absent in healthy samples.
ELISA and PCR were efficient in diagnosis of viruses in suckers and micropropagated cultures.
