INDUCTION OF CALLUS FROM HYPOCOTYLEDONS AND COTYLEDONARY LEAVES OF CERATONIA SILIQUA

Abstract
Pale yellow callus was formed from segments of seedlings on MS medium + 2,4-D (3 mg.L-1) + BAP (3 mg.L-1) after 4 weeks by 58.3%. No callus was induced in case of using MS medium + NAA (0.5 mg.L-1) + Kin (1 mg.L-1) and MS hormone-free.
On the other hand, callus was formed on MS medium+ BAP (2 mg.L-1) by 71.4%. A large amount of callus formed on MS medium + TDZ (0.5 mg.L-1) and the callus regenerated small leaflets structures when subcultured on MS + TDZ (1 mg.L-1) after 4 weeks.
For cotyledonary leaves, callus was formed on MS + NAA (0.5 mg.L-1) + Kin (1 mg.L-1) and MS + 2,4 D (3 mg.L-1) + BAP (3 mg.L-1) by 25 and 100%, respectively.
No callus was induced in case of using MS hormone-free.
Different concentrations of TDZ were also applied, for increasing and multiplication of the callus from cotyledonry leaves.
