THE EFFECT OF NAA AND KIN ON THE INDUCTION OF CALLUS FROM RETAMA RAETAM PLANTS

Abstract
Different parts of the retama plant (radicals and cotyledons) were placed on MS medium supplemented with different concentrations of NAA and Kin.
White compact callus was formed from radicals on MS medium plus (5 mg/L) + Kin
(2.5 mg/L). When MS medium plus (4.0 mg/L) + Kin (1.0 mg/L) was used, a small structure-like bud was formed after three months of culturing.
When that bud was sub-cultured on MS medium plus NAA (1.0 mg/L) + Kin (1.0 mg/L), no further growth was observed.
The highest percentage for callus formation (33.3%) from cotyledon explants was obtained on MS medium plus NAA (4.0 mg/L) + Kin
(0.5 mg/L) and on MS medium plus NAA (5.0 mg/L) + Kin (2.5 mg/L) after four weeks.
The effect of auxin to cytokinin ratio on multiplication of previously induced callus was studied.
The produced callus was sub-cultured on MS medium plus NAA (2.5 mg/L) + Kin (2.5 mg/L) and on MS medium plus NAA (1.5 mg/L) + Kin
(3.5 mg/L). The response percentages were 41.66% and 91.66% respectively.
