EFFECT OF CHITOSAN AND JASMONIC ACID RELATED COMPOUNDS ON SHOOT FORMATION FROM RHIZOME CULTURES OF AN ORIENTAL CYMBIDIUM SPECIES

Abstract
We examined the effect of a jasmonic acid related compound (n-propyl dihydrojasmonate; PDJ), methl jasmonate (Me-JA), and chitosan H added to modified MS medium on shoot formation of a Cymbidium species (C. kanran Makino). Single addition of Me-JA was effective for shoot formation from rhizome cultures.
Combination treatments of chitosan H with Me-JA were also effective for shoot formation from rhizome cultures.
Percentage of rooting was highest (64.3%) in a combination treatment of 1 mg L-1 chitosan H + 0.01 µM Me-JA. Single addition of PDJ to culture medium was effective for shoot formation from rhizome cultures, and showed a higher number and rate at 0.1 µM PDJ. Combinations of Chitosan H with PDJ were more effective for promoting shoot formation from rhizome cultures.
The addition of PDJ in combination with 1 mg L-1 chitosan H was most effective for shoot formation.
Rooting from newly formed shoots was promoted by addition of 1 mg L-1 chitosan H. The percentage of rooting was higher (60%) in the combination treatment of 1 mg L-1 chitosan H with 0.01 µM PDJ. These results indicate that a combination of jasmonic acid derivatives and chitosan has a significant effect on both shoot and root formation from cultures of oriental Cymbidium species (C. kanran).
