STUDIES ON IN VITRO MULTIPLICATION OF CARNATIONS I. THE INFLUENCE OF SOME CYTOKININS ON THE DIFFERENTIATION OF SHOOT APICES /RESEARCH NOTE 1/
The first one – on the influence of kinetin, BA and zeatin riboside in concentrations 1, 2, 4, 8 and 16 /uM on the growth and development of shoot apices, and second one – on the influence of transplantation from media with BA /3,6 and 18 /uM to the media with kinetin /4 /uM/ on the same phenomena.
It was found that culturing shoot apices 20 days on the medium with 6 /uM BA and transplanting them to the medium with 4 /uM kinetin is the best regarding good multiplantlet formation.
One of the most widespread methods of micropropagation is multiplication based on multiplantlets induction from shoot apices.
Multiplantlets can be divided and single shoots further multiplied on similar media.
The medium inducing formation and growth of adventitious shoots from shoot apices should contain more cytokinin in relation to auxins.
This regularity isn’t relative to carnation only but to other species as well /Haramaki 1971, Fujino et al. 1973, Murashige et al. 1974, Gregorini et al. 1976/.
In the case of carnation the influence of cytokinins on adventitious shoot formation was investigated by some authors /Hoof 1971, Pennazio 1975, Davis et al. 1977, Hempel 1977/. Hoof /1971/ was the first to observe this phenomenon.
He used in his experiments kinetin and BA. He found that during the growth of shoot apices on media containing 0.5 mg/l kinetin and 0.1 mg/l BA adventitious shoots were formed.
Pennazio /1975/ examined the influence of kinetin on the growth of shoot apices and obtained an increase of lateral shoot formation correlated with an increase of kinetin concentration in the media /from 0.001 to 1.0 mg/l/. Also Earle and Langhans /1975/ obtained good multiplantlet formation on the medium containing 0.5 mg/l kinetin.
Davis et al. /1977/ compared the influence of some cytokinins on multiplantlet formation.
They obtained the best results on kinetin rich media in comparison to media with BA, zeatin and DMAAP. On the media with kinetin the number of shoots formed as well as their quality /lack of deformation were satisfactory.
