ORGANOGENESIS AND SOMATIC EMBRYOGENESIS FROM CALLUS OF WINGED BEAN (PSOPHOCARPUS TETRAGONOLOBUS (L.) DC.)

Callus tissue and suspension cultures of several cultivars of winged bean, Psophocarpus tetragonolobus (L.) DC, were established from hypocotyls, cotyledons, and epicotyl explants.
Callus growth was observed best in Murashige-Skoog (MS) salt-sucrose culture medium supplemented with 1 mg/l each of 2,4-dichlorophenoxyacetic acid (2,4-D) and naphthaleneacetic acid (NAA) and 1 mg/l each of NAA and kinetin or 6-benzyladenine (BA). Callus clumps when transferred to a MS medium supplemented with 1 mg/l BA with or without kinetin produced shoots and upon transfer to a medium with 1 mg/l indoleacetic acid (IAA) produced roots to give plantlets.

Somatic embryogenesis was obtained by transferring newly initiated calli maintained in a medium with 0.1 – 5.0 mg/l 2,4-D, 2 mg/l each of 2,4-D and NAA or 5 mg/l NAA to a medium free of hormones, supplemented with full or half strength MS salts.
Three varieties of winged bean, TPt-1, TPt-2 and UPS-122 responded to form somatic embryos or embryo-like structures in liquid suspension cultures.
