Abstract
Isolation of a leaf from the mother plant or a cut in the midrib of the intact leaf was found to trigger the growth of dormant epiphyllous buds in Kalanchoe mortagei. However, the application of CoCl2 immediately to the cut inhibited the outgrowth of dormant buds.
Transfer experiments involving CoCl2 and cycloheximide demonstrated that ethylene and new proteins mandatory for bud outgrowth are synthesized within the first 5 and 18 h of leaf isolation respectively.
Pulse chase experiments using (14C) serine also substantiated this finding of de novo synthesis of a specific protein (IAA oxidase). This enzyme exhibited tremendous augmentation in its level during the first 24 h followed by a declining trend.
