Abstract
Young floral buds of the normal and male sterile stamenless-2 (sl-2/ sl-2) mutant of tomato were cultured successfully in vitro in modified Murashige and Skoog medium.
The normal buds required benzylaminopurine (BAP), whereas the mutant buds also needed gibberellic acid (GA3) for growth and development.
Naphthaleneacetic acid (NAA) was inhibitory to the growth of all floral organs, and abscisic acid (ABA) and 2-chloroethyltrimethyl ammonium chloride (CCC) specifically inhibited the growth of stamens of normal floral buds.
The mutant buds grew to maturity in 4 to 5% sucrose; concentrations which were inhibitory to the normal buds.
The observations reported suggest that the sl-2/sl-2 mutation affects the endogenous gibberellins which through lower levels of soluble sugars, affect the development of pollen grains.
