INFLUENCE OF LIGHT INTENSITY ON TOMATO CALLUS PRODUCTION

Abstract
Tomato leaf disc explants were grown on semi-solid nutrient media under photon flux densities which range from darkness up to a level which was apparently inhibitory to callus tissue production and subsequent differentiation (190μE/m2/s). Cultures were raised within a controlled environment cabinet but as previous work has indicated tissue ambient temperature increases with increasing photon flux density.
By maintaining culture vessels in separate thermostatically controlled water baths, an attempt was made to separate the effect of irradiance from that of a temperature.
Maximum callus production was found to occur at 70uE/m2/s and at a temperature of 30°C. Maximum callus cell size was found to coincide with the optimum temperature for callus production.
