AGROBACTERIUM-MEDIATED TRANSFORMATION OF APPLE AND STRAWBERRY USING DISARMED TI-BINARY VECTORS

Abstract
The disarmed Ti-binary vector pBIN6 in Agrobacterium tumefaciens has been used to produce transgenic apple and strawberry plants from transformed leaf discs and petioles.
Genes encoding nopaline synthase (nos) and neomycin phosphotransferase (npt II) have been shown to be present in host DNA by Southern blot analyses.
Their expression has been confirmed by enzyme assay and rooting bioassays in the presence of kanamycin.
In the case of strawberry it was possible to perform inheritance studies on the segregation of nos by self-pollinating greenhouse grown plants.
The nos gene segregated 3:1 in in vitro germinated seedlings according to the expected Mendelian ratio for a single dominant.
