THE ROLE OF ABA IN INDUCTION OF ANDROGENESIS IN BARLEY

Abstract
Androgenesis is not a naturally occurring event in angiosperm and only seems to be induced by certain abiotic stress conditions.
As stress treatments are generally associated with ABA signaling, a role for this stress hormone in the reprogramming of pollen development is very important.
Therefore, two barley cultivars with different regeneration efficiency were studied with regard to their endogenous ABA level and responses. Under the same mannitol pretreatment and culture conditions, regeneration efficiency in the barley cultivar (cv.) Igri was about ten times higher than in the cv.
Digger that was only partially reflected by a difference in viable microspores after anther pretreatment.
Under this mannitol pretreatment, for cv.
Igri, a much higher endogenous ABA level was reached as compared to cv.
Digger.
The effect of ABA was to stimulate the increase in percentages of viability and/or to reduce the number of bi-nucleate microspores.
Moreover, in cv.
Igri, direct culture of anthers for 4 days without any pretreatment caused an increased number of bi-nucleate microspores as compared to the microspores with pretreatment for 4 days.
Based on our observations, we suggest that the difference in regeneration efficiency in cv.
Igri and cv.
Digger is related to their difference in endogenous ABA production level under mannitol pretreatment and their responsiveness to ABA. One of the effects of ABA is likely due to reduction of the number of binucleate microspores.
