Abstract
In this study, at first, we tried to induce protocorm-like bodies (PLB) from rhizome-segments of Cymbidium kanran Makino (terrestrial species). In secondary experiments, we succeeded in inducing rhizome from PLB cultures of an epiphytic Cymbidium cultivar.
Addition of paclobutrazol to modified MS medium induced PLB formation from rhizome segment cultures of C. kanran. The combination of benzyladenine (BAP) with paclobutrazol reduced the number of PLB. Application of BAP induced protocorm-like shoot (protocorm-like bulb with short leaves) without root. 10 µM BAP was significantly effective for induction of protocorm-like shoots.
Single addition of 10 µM BAP resulted in increase of shoot formation.
Paclobutrazol in culture medium inhibited BAP-induced leaf formation from rhizome cultures.
Application of GA3 to PLB cultures of epiphytic type Cymbidium resulted formation of rhizome.
GA3 treatment at 100 µM was effective for the rhizome formation from PLB cultures.
Single addition of 10 µM paclobutrazol was effective on proliferation of PLB of the epiphytic Cymbidium . It is supposed that cytokinin and gibberellin have important roles for the ecological differentiation of epiphytic and terrestrial Cymbidium species.
