GENOTYPE EFFECTS ON AGROBACTERIUM MEDIATED GENETIC TRANSFORMATION OF BRASSICA NAPUS

Abstract
Agrobacterium tumefaciens mediated transformation was applied to transfer genes into three genotypes of Brassica napus, namely a spring type oilseed rape cv. ‘Drakkar’, a resynthesised high erucic line ‘RS 306’ and a high erucic line ‘11502’. Plant expression constructs were developed and transferred via Agrobacterium tumefaciens strains GV3101/pMP90 or C58ATHV/pEH101. Transformation rates with etiolated hypocotyls averaged 44 % of the total number of explants for genotype ‘RS 306’ which displayed the highest transformability among the three genotypes tested.
The high level of transformation rate based on the high regeneration ability, with more than 90 % of the explants forming adventitous shoots.
The formation of transformed shoots occurred mainly using the Agrobacterium strain ATHV and the vir inducer acetosyringone.
In contrast shoot regeneration was unsatisfactory with ‘Drakkar’ and line ‘11502’ and did not exceed 21 % of the total number of hypocotyls.
The regeneration efficacy could not be increased by using a medium supplemented with 5 mg/l BAP and 0.5 mg/l NAA.
