EFFECTS OF AGAR IMPURITIES ON IN VITRO PROPAGATION OF RANUNCULUS ASIATICUS L.

Abstract
Analysis of mineral elements in dry agars and in the washing liquid collected from the dialysed agars showed that three agar brands (Oxoid=OX, Merck=MK and Roth=RT) contain different amount of impurities which might be differently available to the tissues.
Elements like Ca, K, Na, Mn and Zn could be removed more easily from RT agar than from OX and MK agars.
Thermal analysis of the different gels suggested that gelled media have plenty of water (ranging between 70% and 90%) and data on liquid collected after gel centrifugation showed that different water availability could be expected for the different gels.
Agar impurities could directly affect the plant metabolism and influence the water availability to the tissues.
The unsatisfactory quality of Ranunculus shoots on RT gels could be overcome by washing the agar and increasing the agar level in the washed gels.
An improved multiplication rate could be only achieved by adding the OX impurities to the washed RT gels.
