EFFECT OF AGROBACTERIUM INCUBATION CONDITIONS ON GENE TRANSFER AFFECIENCY ‘GALA’ AND ‘MCINTOSH WIJCIK’ APPLE

Abstract
With the aim to improve the efficiency of Agrobacterium-mediated transformation in apple cultivars, we investigated some of Agrobacterium incubation conditions including bacterial density in liquid YEB medium and a virulence induction medium and the presence of antibiotics in liquid YEB medium.
For transformation, leaf explants from ‘Gala’ and ‘McIntosh Wijcik’ apples were infected with A. tumefaciens LBA4404 harboring pGA 1532 or pGA 1534 carrying MdMADS2 or MdMADS4, respectively.
The regeneration rate and the transfer efficiency were highest when using Agrobacterium cells of OD600 0.9 to 1.1 incubated at 28℃ for 24 hours in liquid YEB medium.
In other hands, there were no effects of the density of the bacterial cells diluted in a virulence induction medium and the presence of antibiotics in liquid YEB medium.
The presented result implies that the growth stage of Agrobacterium cells is an important factor for improving transfer efficiency in apple cultivars.
