Molecular characterization of local pear cultivars collected across Greece

Abstract
Local cultivars may show resistance to biotic and abiotic stresses and adaptation to the environmental conditions of each region.
Local cultivars of pear grow consistently in arid and dry island regions, without cultivation care, while foreign cultivars often fail and are uprooted.
This material often has important commercial characteristics or may be the necessary primary material for the creation of new cultivars resistant to climate change.
During the “Fruitrees2Safeguard” research program, that took place from 2018 to 2022, 264 different local fruit cultivars/genotypes across 32 different regions across Greece were investigated.
Interestingly, a high number of pear cultivars (95) were recorded (36% of total fruit trees cultivars), with most of them found in the island of Lesvos (16), in Imathia (14), in the island of Ikaria (13) and in the island of Samos (12). Morphological/Phenotypic data were collected according to the instructions of UPOV or CPVO in 30 pear cultivars and samples were collected for further genetic and biochemical analysis.
The genetic variability of 86 pear cultivars was studied using 7 Inter Simple Sequence Repeat (ISSR) markers and a total of 104 bands were generated.
A total of 49.81% of the ISSR fragments were polymorphic and 12 private bands were detected.
Results of Analysis of Molecular Variance (AMOVA) revealed percentages of 34% among populations and 66% within populations.
Also, high percentages of polymorphic loci were found among the pear cultivars collected from 10 different regions, with the highest percentage (60.58%) belonging to cultivars from Samos.
Principal Coordinates Analysis (PCoA) resulted in clustering the pear cultivars in 3 different groups.
These findings could further be utilized in future pear breeding programs for selecting individuals with favorable traits.
