Genetic analysis of several special traits of a semi-wild XIS cucumber (Cucumis sativus var. xishuangbannesis)

Abstract
XIS cucumbers are originated from southwest China, and are characterized by their least tolerance to low temperature, downward female flowers, fewer both male and female flowers, more lateral branches.
The objective of this study was to construct a recombination inbred line (RIL) population and to map the QTLs that control the distinct agronomic traits of XIS cucumbers.
The final RIL population consisted of 152 individuals and showed a wide variation in morphological traits including fruit shape, color of flesh and pericarp, carpel number, tubercule size and density, spine color, and leaf bitterness.
No QTL signal of generation in low temperature was detected in the Bulk Segregation Analysis (BSA), despite of the high quality of phenotypic survey and whole genome resequencing.
This indicates that classic nuclear inheritance may not able to explain the trait of cold germination.
QTLs controlling four flower-related traits were mapped by kompetitive allele-specific PCR (KASP) markers, which are male flower number (MFNum1.1 and MFNum6.2), female flower number (FFNum1.1 and FFNum2.1), female flower downward (FFD3.1), and branching (Branch1.1).
