Searching grapevine wild germplasm for new sources of powdery mildew (Erysiphe necator) resistance

Abstract
Grapevine is one of the most widely cultivated and highest value horticultural crops, due to its use for wine, table grape and raisin production.
The vast majority of cultivated grapes are cultivars of Vitis vinifera L., the European grapevine, which is considered to have the highest fruit quality of the Vitis species.
However, V. vinifera is susceptible to a wide range of pests and diseases.
Powdery mildew (Erysiphe necator) is the most damaging fungal disease in dry and warm areas such as the Mediterranean basin.
Nowadays, disease control relies on massive application of fungicides which have a negative impact in the environment, generate fungal resistance and impose an extra cost to wine growers.
In addition, restrictions on the number of sprayings and levels of residues have increased in European legislation.
In this context, screening for new sources of resistance to fungal diseases is a promising alternative to conventional grapevine pest management in order to increase viticulture sustainability through the use of new resistant cultivars. Ren 1 locus has been previously identified in V. vinifera L. ssp. sylvestris germplasm.
In this work, we have conducted a screening of a nuclear collection of Spanish wild germplasm, (62 accessions), in order to identify potential resistant genotypes.
In vitro assays were carried out on detached leaves with 4 replicates and the degree of infection was evaluated following a 1-5 scale at 7- and 14-days post inoculation with ‘Carignan’ and ‘Kishmish vatkana’ as susceptible and resistant controls, respectively.
Accessions with the best performance were tested at least 3 times to confirm results.
Seven accessions presented reduced mycelium growth and reproduction, and were further genotyped with four molecular markers (SSRs) flanking the Ren1 locus.
None of the accessions carried resistant alleles previously described as linked to the resistance.
Further research will focus on identifying different mechanisms of resistance among those accessions.
