Effects of exogenous spermidine on the ascorbate-glutathione (AsA-GSH) cycle in the roots of Cerasus sachalinensis seedlings under water-logging stress

Abstract
Cherry is known for its sensitivity to soil water-logging.
Heavy summer rains increase the risk of water-logging in Chinese cherry production areas.
However, how to improve the ability of cherry roots to cope with water-logging remains unknown.
Here, using a double-pot method to simulate short-term water-logging conditions, the effect of exogenous spermidine (Spd) on metabolite content and key enzyme activity in the ascorbate-glutathione (AsA-GSH) cycle, and on the content of hydrogen peroxide (H2O2) and malondialdehyde (MDA) in the roots of Cerasus sachalinensis seedlings under water-logging stress were investigated.
The results showed that exogenous Spd decreased the accumulation of MDA and H2O2 in the roots of C. sachalinensis seedlings after 24 h of water-logging exposure compared to the accumulation of MDA and H2O2 in the roots of control plants.
At 72 and 120 h of water-logging, the reduced ascorbic acid (AsA) and reduced glutathione (GSH) content, the dehydroascorbate reductase (DHAR) and monodehydroascorbate reductase (MDHAR) activities, and the ratios of AsA to dehydroascorbate reductase (DHA) and of GSH to oxidized glutathione (GSSG) were all significantly higher in the roots of seedlings treated with exogenous Spd than in control group seedlings.
These results indicated that exogenous Spd can increase the AsA content and accelerate the running rate of the AsA-GSH cycle by enhancing the activities of two key AsA regenerating enzymes (DHAR and MDHAR), thereby increasing the ability of C. sachalinensis seedlings to scavenge MDA and H2O2, and to some extent alleviating water-logging stress injury to the seedlings of C. sachalinensis.
