Construction of an infectious clone of a sweet cherry isolate of hop stunt viroid and identification of its infectivity

Abstract
Hop stunt viroid (HSVd) is a member of the genus Hostuviroid of the family Pospiviroidae that causes serious dapple fruit symptoms in infected sweet cherry.
In order to analyze the pathogenesis mechanism of the sweet cherry HSVd-SC isolate, we constructed the infectious clone named HSVD-PBI-SC1 via the homologous recombination method.
The infectious clone was transformed into Agrobacterium tumefaciens GV3101. The tobacco, cucumber and sweet cherry rootstocks inoculated with Agrobacterium tumefaciens displayed typical symptoms of stunting, leaf curling and malformation.
HSVd was also detected in the inoculated plants by RT-PCR. The successful construction of Agrobacterium-mediated infectious clones of HSVd will contribute to the understanding of the viroid pathogenesis mechanism of HSVd.
