Distribution of abscisic acid in strawberry fruit visualized utilizing MALDI-TOF MSI

Abstract
Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging (MALDI-TOF MSI) is a powerful tool which can visualize the spatial distribution of various compounds in a tissue specimen.
While distribution of soluble carbohydrates, organic acids and anthocyanins in strawberry fruits has been examined utilizing MALDI-TOF MSI, few trails have been done on plant hormones.
It is well known that strawberry fruits contain abscisic acid (ABA), which plays an important role in fruit maturation.
Furthermore, ABA may act as an inhibitor of seed germination in strawberry fruits, as was reported for watermelon fruits.
However, since the concentration of ABA in the fruits is quite low, detecting ABA using MALDI-TOF MSI was not a viable method.
So, we tried to visualize the distribution of ABA in a strawberry fruit following Enomoto’s method, where the specimen was pretreated with Girard’s reagent T (GirT) which can assist the ionization of ABA molecules.
The results showed that the concentration of ABA was high in cortex tissue, and accumulation of ABA was clearly confirmed at the vascular bundle conjugated with achenes.
This finding may lead to understanding of the mechanisms of fruit maturation and the inhibition of seed germination in strawberry fruits.
