Evaluation of Bacillus spp. for biological control of Fusarium stem rot in Anoectochilus formosanus

Abstract
Anoectochilus formosanus Hayata is an orchid rich in bioactive compounds with significant medicinal properties.
The stem rot disease caused by Fusarium oxysporum is a major challenge that can cause significant yield losses of A. formosanus Hayata.
Only a few fungicides and non-chemical agents are available for controlling this disease.
Therefore, this study aims to explore the biological agents for their potential to control Fusarium stem rot.
Several microbial strains were isolated and screened from rhizosphere soil samples of A. formosanus Hayata.
Dual culture assay indicated four isolates, AFA5, AFB4, AFC4, and AFC5, demonstrating significant antagonistic effects against the F. oxysporum mycelial growth, with an inhibition rate above 55.9%. In addition, these isolates could effectively inhibit F. oxysporum spore germination, with AFA5 exhibiting the highest inhibition rate of 97.7%. Using 16S rDNA and gyrB gene sequencing analysis, these four antagonistic bacteria were identified as Bacillus amyloliquefaciens. Moreover, we discovered that these antagonistic bacteria secrete multiple extracellular enzymes, including protease, amylase, cellulase, and lipase, while enhancing phosphate solubilization.
Further, the suspension of B. amyloliquefaciens isolates was directly applied onto A. formosanus Hayata roots before spore inoculation, resulting in a 35.0-43.3% reduction of Fusarium stem rot disease incidence.
In the pot experiments, the treatment group drenched with bacterial suspension effectively demonstrated a 14.6 to 20.8% reduction in A. formosanus Hayata mortality rate.
In conclusion, this study successfully identified four Bacillus amyloliquefaciens strains from rhizosphere soil, exhibiting a significant potential as bio-control agents for Fusarium stem rot disease in Anoectochilus formosanus Hayata.
