Isoprothiolane regulates endogenous abscisic acid and indole-3-acetic acid during grape berry maturation

Abstract
The effects of isoprothiolane (IPT) on endogenous abscisic acid (ABA) and indole-3-acetic acid (IAA) in grape maturation were investigated. ‘Shine Muscat’ (Vitis labruscana Bailey × V. vinifera L.) grape clusters were dipped into 2.75 mM of IPT at 44 days after full bloom (1 week before veraison). IPT treatment significantly decreased sugars (glucose, fructose, and sucrose) concentrations at 8, 15, and 38 days after treatment (DAT). Endogenous ABA concentrations declined significantly with downregulation of VvNCED1, a key biosynthesis gene for ABA, in IPT-treated grape peels at 15 and 38 DAT. Moreover, the treatment lowered the expression levels of the ABA signaling gene (VvABI3). At 8 DAT, the endogenous IAA concentrations were significantly decreased in IPT-treated grape peels, which corroborated the upregulation of IAA-amido synthetase gene (VvGH3-like) expression.
Moreover, the auxin polar transporter gene (VvPIN3-like) was significantly downregulated by IPT treatment at 38 DAT. These results suggested that IPT treatment affects grape maturation by modulating the endogenous levels of both ABA and IAA as well as their related genes.
